灵芝孢子粉对异丙肾上腺素致心肌缺血损伤大鼠apelin表达的影响

摘    要:目的: 观察异丙肾上腺素(ISO)致心肌缺血损伤大鼠血浆和心肌组织apelin的变化,探讨灵芝孢子粉抗心肌缺血损伤的作用机制。方法: 将大鼠分为5组:正常对照组、ISO模型组、灵芝孢子粉高、中、低剂量治疗组,连续灌胃给药7周。皮下注射ISO建立大鼠心肌缺血损伤模型,ELISA分析检测血浆和心肌apelin的含量,半定量RT-PCR方法检测心肌apelin mRNA表达,电镜观察心肌组织病理形态学的变化。结果: 与正常组比较,ISO组大鼠血浆和心肌组织apelin含量均下降,心肌组织中apelin mRNA水平表达下调(P<0.01)。与ISO组比较,灵芝孢子粉治疗组大鼠血浆和心肌组织apelin含量以及心肌apelin mRNA表达水平显著升高(P<0.01或P<0.05),同时,血中NO含量显著增加;心肌病理改变明显减轻。结论: 灵芝孢子粉抗大鼠心肌缺血损伤的机制可能与其上调心肌组织apelin mRNA的表达,提高apelin水平有关。

Abstract:AIM: To investigate the changes of apelin in blood and myocardium, and the protective mechanisms of ganoderma spores on myocardial ischemia injury induced by isoproterenol in rats. METHODS: The model of myocardial ischemia injury was induced by subcutaneous injection of high dose isoproterenol. Enzyme linked immunosorbent assay (ELISA) was used to measure the apelin contents in blood and myocardium. Semi-quantitative RT-PCR was used to measure the apelin mRNA level in myocardium. Electron microscope was used to observe the pathomorphological changes of myocardium. Ganoderma spores was administered i.g. for 7 weeks. RESULTS: Compared with the normal control group, the apelin contents in blood and myocardium and the apelin mRNA level in myocardium were significantly decreased in the model group (P<0.01). Compared with the model group, the apelin contents in blood and myocardium and the apelin mRNA level in myocardium were obviously elevated in the ganoderma spores groups (P<0.01 or P<0.05). Meanwhile, the NO contents in blood and myocardium were also obviously elevated. The pathological damage of myocardium was obviously relieved. CONCLUSION: The results suggest that the protective mechanisms of ganoderma spores on myocardial ischemia injury may be related to the elevation of apelin contents and the mRNA level.
