| 摘 要： | 目的 寻找更好的肾组织单细胞悬液制备方法。方法 以肾缺血再灌注大鼠肾为标本 ,分别采用剪碎法和研磨法制备肾组织悬液。经台盼蓝染色测细胞存活率 ,并进行流式细胞术 (FCM )分析。结果 剪碎法细胞形态完整、存活率高 ;2种方法细胞凋亡水平 (APO)、CV(coefficiencyofvariation)值有显著性差异 (P <0 .0 1)。结论剪碎法制备肾组织单细胞悬液优于研磨法

| Abstract: | ObjectiveTo inves tigate the mechanical methods for processing renal tissue into single cell sus pension.? Methods The samples for experiment were coll ected from rats suffering from renal reperfusion injury (clamping bilateral ren al pedicles for 45 minutes followed by 24 h of reperfusion). The single cell su spension was prepared by using scissors or homogenator. The cell viability was a ssessed by typan blue exclusion; the morphologic changes of cells were obs erved under microscope; and the suspension was analyzed by FCM.?Re sultsMore intact cells and better viability were found in the sciss or-cut preparation than in the homogenated (P<0.01).The apoptosis rate (APO) and coefficiency variation (CV) were lower in the former than in the latter ( P<0.01).?ConclusionCutting with scissors is prefera ble to using homogenator for making single cell suspension from renal tissue. |
