白血病骨髓细胞中p53蛋白的表达及其意义

采用人类特异的p53单克隆抗体pAB1801,用ABC免疫组化法即生物素-抗生物素法对36例白血病骨髓细胞进行了免疫组化检测,结果突变p53蛋白表达阳性率为55.56％。p53突变与白血病的类型有重要关系:在急性粒细胞白血病中p53有较高表达,在急性淋巴细胞白血病中p53表达较低,在慢性粒细胞白血病的慢性期突变p53的表达很低,而急变期可出现类似急性粒细胞白血病的突变p53基因表达。研究结果提示p53蛋白表达有可能作为慢性粒细胞白血病急变期的早期诊断指标之一。

The Expression of p53 Protein in Leukemic Bone Marrow Cells and Its Clinical Significance

ABC immunohistochemical studies of protein expression were performed on bone marrow samples of 36 cases with different types of acute or chronic leukemias using anti - p53 monoclonal antibody pAB1801. The positive rate of mutational p53 protein expression was 55.56% in 36 leukemia cases. There was a close relationship between p53 positive rate and the type of leukemias. There was higher p53 protein expression in ANLL while lower expression in ALL. During the chronic phase of CML, the expression of p53 protein was very low, but once a patient got into the blast crisis phase, the p53 protein expression became very high. This finding demonstrated that expression of p53 protein could be one of the early diagnostic indications of the blast crisis of CML.
