摘    要:目的探讨凋亡相关基因聚集素(clusterin)在前列腺癌不同周期时相的差异表达及意义。方法应用改良的胸腺嘧啶核苷(TdR)和高压笑气双阻断法将前列腺癌细胞系PC-3同步在不同的周期时相,应用流式细胞术检测同步化的效率。应用RT-PCR及Western blotting方法检测不同周期时相聚集素在mRNA和蛋白水平的表达。结果前列腺癌细胞系PC-3的M、G1、S和G2期细胞同步化效率分别为92.1%、87.0%、80.2%和75,9%;聚集素在不同周期时相均有表达且存在差异,G1和S期表达水平较低,M期和G2期表达水平较高,RNA和蛋白表达具有一致性。结论细胞周期对聚集素的表达有较大的影响,对于采用聚集素途径治疗前列腺癌具有指导意义。

Abstract:Objective To investigate the differential expression of apoptosis associated gene,clusterin,throughout the cell cycle and its possible clinical significance.Methods The prostate cancer cell line PC-3 was synchronized in M,G_1,S and G_2 phases using modified thymine deoxyriboside(TdR) and high pressure N_2O techniques.The efficiency of synchronization was detected by flow-cytometry.RT-PCR and Western blotting methods were used to examine the expression of clusterin in different phases throughout the cell cycle at the mRNA and protein levels.Results The synchronization rates of M,G_1,S and G_2 phases were 92.1%,87.0%,80.2% and 75.9%,respectively.Clusterin was constitutively expressed throughout the cell cycle,but both the mRNA and protein expression levels of G_1 and S phases were lower than those of M and G_2 phases.Conclusions Cell cycle can influence the expression level of clusterin significantly,which may have certain clinical implication for the treatment of prostate cancer.
