REM睡眠剥夺后大鼠脑组织氨基酸类神经递质含量的变化

摘    要:目的:观察睡眠剥夺(sleep deprivation,SD)对大鼠脑组织γ-氨基丁酸(GABA)、谷氨酸(GLU)含量的影响。方法:用小平台水环境法(flower pot technique)对大鼠进行连续96h快速动眼相(rapid eye movement,REM)睡眠剥夺,以正常饲养组及大平台组为对照,高效液相色谱法测定睡眠剥夺后大鼠额叶皮质、脑干、下丘脑等脑区GABA、GLU含量的变化。结果:睡眠剥夺后大鼠各脑区脑组织GABA含量。GABA/GLU值均较对照组明显增高(P<0.01),而GLU含量在脑干、下丘脑两脑区亦较对照组明显增高(P<0.05)。结论:睡眠剥夺导致大鼠脑组织氨基酸类神经递质含量及比例的改变,是睡眠剥夺过程的神经生物学机制之一。

Abstract:Objective To investigate the effects of REM sleep deprivation on GABA and GLU contents in the rat brain. Methods Animals were randomly divided into three groups:REM sleep deprivation group (small platforms 6.5 cm in diameter,SD) ,tank control group (large platforms 14 cm in diameter, TC)and cage control group(CC). The rats of SD group were deprived of REM sleep for 96 hours by flower pot technique. High performance liquid chromatography(HPLC) was used to determine the contents of GABA and GLU in three brain regions of the rats: frontal cortex,hypothalamus and brain stem. Results SD group showed the increase in contents of GABA as well as ratio of GABA/GLU in all the three brain regions ( P<0.01)compared to CC group and GLU contents were also the increased in the brain stem and hypothalamus of SD group ( P<0.005). Insignificant changes in GABA and GLU contents of the three brain regions were found in TC group. Conclusion REM sleep deprivation induces significant changes in the contents and proportion of the amino - acid neurotransmitters in the rat brain.
