摘    要:研究了1985年至1986年本所繁育场发生的3次较大规模Wistar乳鼠流行性腹泻,经过电镜、聚丙烯酰胺凝胶电泳(PAGE)、实验感染动物、血清学及荧光组化抗原定位,证实病原为B组轮状病毒(GBRV),且与我国发现的成人腹泻轮状病毒(ADRV)有交叉反应,其RNA电泳型与国外近来发现的唯-一株大鼠B组轮状病毒有差异,同时观察了病理学变化。

Abstract:During 1985-1990, three outbreaks of diarrhea in infant Wistar rats were studied. The etiologic agent was identified to be group B rotavirus by means of EM, PAGE and ELISA. Two kinds of virus particles were found in feeal specimens. The complete particle surrounded by double membrane was 70nm in diameter, and the incomplete particle without outer membrane was 50nm in diameter. The genome from the virus had 11 bands of RNA fragmant in PAGE and the distrbution pattern of 4-2-1-1-1-1-1 was different from the typical pattern 4-2-3-2 of group A rotavirus. The positive detection rate was 67% and 93% by EM and PAGE, respectively. The new rotavirus had no serologic reaction with group A rotavirus (the tyrical rotavirus SA-11), but had a strong reaction with group B rotavirus by ELISA. Wistar and SD suckling rats suffered from acute diarrhea after experimental infection by oral inoculation for 24 hrs, and the same virus was detected. The pathological changes showed vacuolation of intestinal epithelial cells and the early presence of cytoplasma eosinophilic material-like inclusion bodies. The virus antigen was detected in intestinal epithelial cells by immunofluorescent histochemical method. Clumps or sporadic viruses were observed in intestinal epithelial cells by EM, and high-electron reticular materials were shown in plasma instead of in organellaes. Rat rotavis (IDIR strain) was found in 1984, and was related to local human diarrhea. We had compared the new rotavirus, IDIR and adult diarrhea rotavirus(ADRV) by PAGE and found that the new rotavirus was more close to ADRV than to IDIR strain. Group B rotavirus from animals was detected for the first time in China.
