基于正交设计大鼠脑动脉血管分叉冠状冰冻切片制备方法研究

摘    要:目的 针对大鼠脑血管分叉部冠状切面,优化冰冻切片的实验条件. 方法 选择冷冻温度、切片厚度和液氮表面速冻时间三因素、三水平设计正交实验,将实验结果输入SPSS 13.0软件,分析得到正交实验方差分析表和单因素统计表,并对实验数据进行统计分析,得到实验的优化条件. 结果 大鼠脑血管分叉部冠状面冰冻切片的最优条件是:冷冻温度-25℃,切片厚度4μm,液氮表面速冻时间15 s. 结论 应用SPSS 13.0软件进行正交实验设计和分析,可以快速获得大鼠脑血管分叉部冠状面冰冻切片优化实验条件,节省实验标本制备和人工劳动成本.

Abstract:Objective To optimize the conditions for preparing the rat cerebral artery bifurcation coronal frozen sections.Methods Three-factor and three-level orthogonal design was processed with SPSS 13.0.Three factors were frozen temperature,section thickness and quick-freezing time above liquid nitrogen.The data from orthogonal design mean square chart and single factor charts were collected and analyzed.Results The optimized conditions for rat cerebral artery bifurcation coronal frozen section were frozen temperature at-25 ℃,section thickness of 4 μm and quick-freezing time above liquid nitrogen of 15 s.Conclusion The optimized experiment factors could be quickly obtained for rat cerebral artery bifurcation coronal section by SPSS13.0.
