醒脑静对高血氨大鼠脑内CREB表达的影响

摘    要:目的:探讨醒脑静改善高血氨大鼠空间学习记忆功能的分子机制。方法:雄性SD大鼠36只,随机分为3组,每组12只:正常对照组(A组),高血氨模型组(B组),醒脑静治疗组(C组),Morris水迷宫观察动物空间学习记忆的变化。大鼠处死后,检测血氨,RT-qPCR检测大鼠脑组织CREB(cAMP应答元件结合蛋白,cAMP-response element binding protein)基因的表达,western blot检测脑组织磷酸化CREB蛋白的表达。结果:与正常对照组比较,高血氨模型组及醒脑静治疗组大鼠血氨水平明显升高。与高血氨模型组相比,醒脑静治疗组大鼠寻找障台的平均逃避潜伏期、游泳总距离均缩短。与正常组比较,高血氨模型组大鼠皮层与海马组织CREB基因mRNA和蛋白表达均明显下降。醒脑静治疗组大鼠皮层与海马CREB基因mRNA和蛋白表达,较高血氨模型组均明显升高。结论:醒脑静在一定程度上改善了大鼠的空间学习记忆能力,其影响脑内CREB表达可能是其发挥作用的重要途径。

Effect of Xingnaojing on CREB Expression in Brain of Rats with Hyperammonemia

Abstract:Objective : To investigate the molecular mechanism of Xingnaojing improving spatial learning - and - memo- ry ability in rats with hyperammonemia. Methods : Thirty - six clean - grade SD ( Sprague Dawley) male rats were divided into three groups randomly:A group (normal control, n = 12) and B group (hyperammonemia model, n = 12) and C group ( treated by Xingnaojing injection,n = 12). And then the rats were observed the variation of spatial learning - and - mem- ory through Morris water maze. After the rats were killed, blood ammonia of rats were tested, and their brain tissue' s CREB (cAMP response element binding protein, cAMP - response element binding protein) gene expressions were detec- ted by RT - qPCR and phosphorylated CREB protein of brain tissue was detected by western blot. Results : Compared with rats of normal control group, the ones in two other groups had higher level of blood ammonia obviously. And compared with rats of hyperammonemia in model group,the ones treated by Xingnaojing showed evidently improved performance of the test. Compared with the nornml group, CREB mRNA and protein expressions were significantly decreased in cortex and hippocampus of hyperammonemia model group rats. And compared with hyperammonemia in model group, CREB mR- NA and protein expressions were significantly increased in cortex and hippocampus of rats treated by Xingnaojing injec- tion. Conclu*ion:Xingnaojing improved spatial learning - and - memory ability in rats to some extent and its effects on CREB expression in the brain may be an important way of its functioning.
