温莪术对大鼠肾间质纤维化的保护作用及其机制研究

目的:研究温莪术油对单侧输尿管(Unilateral Ureteral Obstruction,UUO)大鼠肾间质纤维化的保护作用及其机制。方法:将40只雄性SD大鼠随机分为假手术组、模型组,莪术组和氯沙坦组。对模型组和治疗组行单侧输尿管结扎术。术后14 d处死大鼠,留取梗阻侧肾组织观察病理损害和间质纤维化程度,免疫组化检测TGF-β1、CTGF在肾组织中的表达。结果:模型组大鼠肾间质纤维化程度最明显,药物干预后肾间质纤维化程度减轻(P<0.05)。免疫组化结果表明模型组大鼠TGF-β1、CTGF表达明显增加;药物干预后TGF-β1、CTGF表达明显低于模型组(P<0.05)。温莪术组与氯沙坦组各指标比较差异均无统计学意义。结论:温莪术可能是通过减少TGF-β1的表达来抑制CTGF的分泌,进而减轻由此导致的肾间质纤维化的损伤。

Protective Effect of Zedoary Turmeric against Renal Interstitial Fibrosis in Rat Model and Its Mechanism

Objective：To investigate the protective effects of Zedoary Turmeric on renal fibrosis of unilateral ureteral obstruction（UUO） rats and its mechanism. Methods ： Forty male Sprague - Dawley （ SD ） rats were randomly subdivided into a sham - operated group, a model group, Zedoary Turmeric group and Losartan treated group. UUO model was induced by ligating the left ureter of rats. All rats were sacrificed 14 days after UUO. Pathological changes of the renal tissue were observed,the protein expressions of TGF -β1 and CTGF were detected by immnnohistochemical staining. Results ： Compared with UUO group, renal fibrosis was attenuated by treatment group （ P 〈 0. 05 ）. Compared with control group,expressions of TGF-β1 and CTGF were significantly up -regulated in UUO group（ P 〈 0.05 ）. In contrast, drug treatment siguificantly down - regulated the expressions of TGF -β1 and CTGF in renal tissue（ P 〈 0.05 ）. No significant difference in effect on indexes was observed between Zedoary Turmeric group and Losartan treated group. Conclusion： Zedoary Turmeric may protect the kidney by down - regulating the expression of TGF -β1 and CTGF in progression of renal interstitial fibrosis.
