人参皂苷Rg3联合顺铂对荷卵巢癌裸鼠移植瘤模型作用研究

摘    要:目的:观察人参皂苷Rg3联合顺铂对荷高转移人卵巢癌细胞系HO-8910PM转移瘤裸鼠的抑制作用。方法:移植瘤裸鼠随机分成6组:荷瘤空白对照组(A组);不同Rg3浓度药物组:2.5 mg/kg(B组)、5 mg/kg(C组)和10 mg/kg Rg3(D组);Rg3和顺铂联用组(E组);顺铂阳性对照组(F组)。分别从裸鼠体重、转移瘤体积变化及相对肿瘤增殖率、淋巴细胞转化实验和NK细胞活性检测观察各组情况。结果:给药6天后,F组裸鼠体重呈显著下降(P〈0.05),E组体重呈极显著下降(P〈0.01)。E组给药6天后,荷瘤裸鼠的瘤体积显著下降,作用明显强于Rg3 C组和F组。D组12天后与A组比较瘤体积显著下降。Rg3 5 mg/kg腹腔注射给药对小鼠脾细胞NK活性有显著的增强作用(P〈0.05)。各剂量Rg3腹腔注射给药对LPS诱导的小鼠B淋巴细胞增殖能力无显著影响(P〉0.05)。结论:人参皂苷Rg3联合顺铂作用于荷卵巢癌裸鼠比两者单独用药具有更强的抑制肿瘤的作用。一定浓度的Rg3能刺激荷瘤裸鼠NK细胞的活性,提高机体免疫功能。卵巢癌应用化疗的同时联合应用Rg3可能会增效解毒、保护机体免疫功能的作用。

Effect of Ginsenoside Rg3 Combined with Cisplan on Transplanted Human High- Metastasis Ovarian Cancer Cell Line H0-8910PM in Nude Mice

Abstract:Objective : To observe the inhibition of ginsengside Rg3 combined cisplatin for nude mice with high - metas- tasis human ovarian cancer cell line H0 -8910PM. Methods:Nude mice were randomly divided into six groups:tumor - bearing control group ( A group) ; different concentrations of drugs Rg3 group: 2.5 mg/kg ( B group ) , 5 mg/kg ( C group) and 10 mg/kg Rg3 (D group) ;Rg3 and cisplatin group (E) ;Cisplatin group (F). The inhibitions of Rg3 and cisplatin were observed from the weight and metastasis tumor volume change of nude mice. The relative tumor proliferation rate,lymphocyte transformation test and NK cell activity were observed in each group. Results : The body weights of group F and group E were both significantly decreased (P 〈 0.05 ) after 6 days of administration. The nude tumor volumes of group E decreased significantly than that of group C and group F when administered for 6 days. The tumor volume of group D decreased obviously compared with that of group A after 12 day's administration. The spleen NK cells activity of Group C were enhanced (P 〈 0.05 ). But there wasnt any change about B lymphocytes in mice with any dose Rg3 (P 〉 0.05). Conclusions : Ginsenoside Rg3 combined with cisplatin has a stronger role in tumor suppression than either drug alone in nude mice with ovarian cancer. Certain concentration of Rg3 can stimulate tumor - bearing nude mice NK cell activity and improve their immune function. Chemotherapy combined with Rg3 may be decrease detoxification and protect the immune function of ovarian cancer patients.
