血浆中miR-31水平与乳腺癌关系的研究

摘    要:目的:探讨血浆中miR-31表达水平与乳腺癌临床病理特征之间的关系。方法:收集2010年12月至2011年3月期间我科符合入选条件的31例女性患者(21例乳腺癌患者,10例非乳腺癌患者)的血液标本,分离血浆并提取其中的miR-31,通过实时荧光定量PCR的方法检测血浆中miR-31表达水平,分析其与乳腺癌临床病理特征间的关系。结果:乳腺癌患者血浆中miR-31的表达水平低于非乳腺癌患者,但差异无统计学意义(P>0.05);淋巴结转移阳性的乳腺癌患者血浆中miR-31的表达水平高于淋巴结转移阴性的乳腺癌患者,但差异无统计学意义(P>0.05);Ⅰ和Ⅱ期乳腺癌患者血浆中miR-31的表达水平低于Ⅲ期乳腺癌患者,但差异无统计学意义(P>0.05);直径小于等于2 cm的乳腺癌患者血浆中miR-31表达水平低于大于2 cm的乳腺癌患者,差异有统计学意义(P<0.05);与ER、PR阴性的乳腺癌患者相比,ER、PR阳性的患者血浆中miR-31的表达水平下调,但差异无统计学意义(P>0.05)。结论:血浆中miR-31的表达水平与乳腺癌的肿瘤大小密切相关,血浆中的miR-31有望成为新一代的乳腺癌的肿瘤标志物。

Abstract:Objective: To investigate the expression of miR-31 in plasm of patient with breast cancer and explore the possibility of miR-31 in plasm as a marker of beast cancer.Methods: Thirty-one cases(breast cancer 21 cases,non-breast cancer 10 cases) registered in thyroid and breast surgery department of No.1 People’s Hospital of Huai’an from December 2010 to March 2011 according to the selected conditions were enrolled.Five milliliters anticogulant blood were sampled to extract plasm and isolate miRNA from plasm respectively.The expression level of miR-31 in plasm were measured by real-time quantitative polymerase chain reaction(real-time qPCR)analysis.The relationship between the expression level of miR-31 in plasm and the fearutre of clinical pathology was analyzed.Results: Compared with non-breast cancer,the expression level of miR-31 in plasm of patient with breast cancer was up-regulated,but there was no significant difference between them(P>0.05).The expression level of miR-31 was up-regulated in lymph positive breast cancer group compared with lymph negative breast cancer,but there was no significant difference between them(P>0.05).The expression level of miR-31 was down-regulated in tumor diameter equal or lesser than 2 centimeter compared with greater than 2 centimeter(P<0.05).The expression level of miR-31 was down-regulated in stageⅠ and Ⅱ breast cancer compared with stage Ⅲ breast cancer,but there was no significant difference between them(P>0.05).The expression level of miR-31 was down-regulated in plasm of patient with ER and PR positive breast cancer compared with ER and PR negative breast cancer,but there was no significant difference between them(P>0.05).Conclusion: This study demonstrates that the expression of miR-31 have a relationship with tumor size and could be used as potential marker of breast cancer.
