姬松茸菌孢多糖对胃癌大鼠IL-2、NK细胞的调节及抑瘤作用

摘    要:目的 探讨中药姬松茸菌孢多糖对胃癌大鼠IL-2、NK细胞的调节及抑瘤作用。方法 将Wistar大鼠 随机分为5组,除空白对照组外,其余4组均经免疫抑制处理(注射阿糖胞苷与60Co全身照射),皮下接种胃癌细胞 悬液,制成胃癌大鼠模型。其中3组分别用姬松茸菌孢多糖大、小剂量及香菇多糖灌胃治疗,正常对照组和模型对照 组分别用相等剂量的生理盐水灌胃。10 d后,观察各实验组肿瘤生长情况及相关免疫指标IL-2、NK细胞的含量或 活性。结果 胃癌大鼠模型组与正常对照组比较,IL-2含量及NK细胞杀伤活性均显著降低(P<0.01)。用药灌胃 后,IL-2含量及NK细胞杀伤活性均显著提高(P<0.01),瘤块明显减小(P<0.01);姬松茸菌孢多糖组的治疗效果 优于香菇组(P<0.05)。结论 姬松茸菌孢多糖有促进胃癌大鼠IL-2、NK细胞免疫功能及抑制肿瘤生长的作用。

Abstract:Objective To explore the effect of polysaccharide from mycelia and spores of Agaricus blazei Murill on IL-2, NK of gastric cancer rat and the antitumor mechanism. Methods The Wistar rats were randomly divided into five groups. Except for normal control group, the other four groups were injected Arac and radiated with 60Co, then transplanted with BGC-823 gastric cancer cells to form gastric cancer rat models. Three groups of which were treated separately by Lentinus edodes polysaccharides (LP), polysaccharide from mycelia and spores of Agaricus blazei Murill (MP) high dose and low dose. The normal control group and the model control group were gived NS. Ten days later, we observed the activity or content of IL-2, NK and the growth of tumor in each experimental group. Results The content of IL-2, and the activity of NK decreased obviously in tumor-bearing animals (P<0.01). After treating, the IL-2 and the activity of NK increased obviously than that of model control group (P<0.01 or P<0.05). The effect of high-dose MP group is more obvious than LP group (P<0.05). Conclnsion MP has positive immune regulatory effect on IL-2, NK of gastric cancer rats and tumor inhibiting effect.
