急性脑外伤组织双向凝胶电泳分析

摘    要:目的利用蛋白质组学方法,寻找急性脑外伤组织的特异蛋白质。方法提取脑外伤患者伤灶组织及正常组织的总蛋白质,通过双向凝胶电泳分离蛋白质,经生物质谱鉴定由图像分析软件所得出的具有表达差异的蛋白质点。结果2组蛋白斑点总体分布相似,主要分布在等电点p13—8,分子量Mr14.4—75kD,分析发现脑外伤组与正常组相比,有18个蛋白质上调和20个蛋白质下调,进一步质谱鉴定,其中7个蛋白质分别为NSFL1辅助因子、硫氧还蛋白、泛素羧基末端水解酶同工酶L1、血小板活化因子乙酰基水解酶IB—r亚单位、谷氨酸脱氢酶前体、二氢嘧啶酶相关蛋白2和微管蛋白。结论脑外伤组与正常组相比蛋白质的表达具有差异。推测,这些蛋白质的变化可能与脑外伤的发展变化具有密切的关系。

Abstract:Objective To find the special protein of acute brain trauma tissue by proteome method. Methods Total proteins of brain trauma tissue and normal brain tissue were extracted separately, then were separated by two dimensional gel electrophoresis and the differently expressed protein spots were identified with biospectrometry. Results The distributions of two group of protein spots were similar, proteins were mainly displayed at the range of pI 3 -8, Mr 14. 4 -75kD. Contrast with normal group, 18 spots increased and 20 spots decreased in the trauma group. Further study indicated that seven of them were NSFL1 cofactor p47, thioredoxin, Ubiquitin carboxyl-terminal hydrolase isozyme L1, Platelet-acfivating factor acetylhydrolase IB gamma subunit, glutamate dehydrogenase, dihydropyrimidinase-related protein 2 and tubulin beta 5. Conclusion There were some differences of proteins expression between normal group and trauma group. The changes of proteins may be tightly correlated with the development of brain trauma.
