| 摘 要： | 目的:探讨P21/waf1/cip1在U937细胞分化中表达情况.方法:用佛波脂(PMA)诱导U937细胞向单核-巨噬细胞分化,经流式细胞术测定细胞P21/waf1/cip1基因表达及细胞周期的变化.用倒置显微镜观察细胞分化的形态学变化.结果:U937细胞向单核-巨噬细胞分化过程中细胞由分散和悬浮转变为粘附和贴壁,呈现了P21/waf1/cip1高度表达,同时细胞周期停滞于G1和G2/M期,无细胞凋亡. 结论:P21/waf1/cip1在U937细胞分化过程中有高表达,并伴随细胞周期变化.

| Abstract: | Objective:To research P 21/waf1/cip1 expression during U937 differentiation. Methods:Induce U937 differentiation towoard monocyte macrophage with PMA, use flow cytometry to assay P 21/waf1/cip1 expression and cell cycle change, and then observe cellular alteration with invert microscope. Results:During cell differentiation, divergent and suspend cell became adhesive and stick with P 21/WAF1/cip1 high express ,cell cycle was arrested in G 1 phase and G 2 M phase without appearence of apopotosis. Conclusion:P 21/waf1/cip1 appears highly expression while U937 differentiation ,and cell cycle was changed with cell cycles. |
