高脂饲料加速阿霉素肾小球硬化模型的建立

摘    要:目的建立局灶节段性肾小球硬化动物模型。方法对SD大鼠行右肾摘除术,分2次尾静脉注射阿霉素,每次3mg/kg,并喂饲高脂饲料。结果该模型表现为浮肿、大量蛋白尿、低蛋白血症、高脂血症;病理学检查、计算机图像分析及免疫组织化学染色显示,肾小球主要表现为系膜基质增加,肾小球出现局灶节段性硬化,细胞外基质FN、CoLⅣ表达上调。结论本模型为研究慢性肾小球疾病的机制及治疗创造了条件。

Abstract:OBJECTIVE To establish animal models for focal segmental glomerulosclerosis.METHOD Nephrectomy of the right kidney of SD rats was performed,followed by caudal vein injection of adriamycin twice(3mg/kg each time) and feeding of high lipid fodder.RESULT This model is manifested by edema, large amount of albuminuria,hypoproteinemia and hyperlipemia.Pathological examinations,computer image analysis and immunohistochemical staining suggested that the glomerule was manifested by the increase of the mesenterium base,focal segmental glomerulosclerosis,and an increase in the expression of extracellular matrix FN and Col IV.CONCLUSION This model provides the necessary conditions for the study of the pathogenesis of chronic diseases of the glomerule.
