蜂胶水提液改善小肠缺血-再灌注大鼠肠系膜微循环

摘    要:目的研究蜂胶水提液(WEP)对小肠缺血-再灌注(I-R)大鼠肠系膜微循环的影响,探讨其减轻I-R损伤的机制。方法40只健康雄性Wistar大鼠随机分为假手术(Sham)、I-R和WEP(50、100、200mg/kg)组,以无创动脉夹夹闭肠系膜上动脉建立小肠I-R损伤模型,于再灌注2h时观测肠系膜微循环变化;分别采用Nackel氏分度法和测定小肠湿/干重比来判定肠道出血损伤和水肿情况,常规制备病理切片光镜下观察肠组织病理学改变;试剂盒测定肠组织超氧化物歧化酶(SOD)活性和丙二醛(MDA)含量。结果(1)与I—R组比较,WEP组小肠病理变化明显减轻,Nackel氏分度和小肠湿/干重比明显降低(P〈0.01);(2)WEP明显改善肠系膜微循环血流状态,呈剂量依赖性地减轻I—R所致的微静脉血流速度减慢(P〈0.05)和白细胞贴壁黏附(P〈0.01),并上调肠组织SOD活性(P〈0.05),抑制MDA生成(P〈0.05)。结论蜂胶水提液通过改善微循环减轻小肠I—R损伤。

Abstract:Objective To investigate the mechanisms underlying mitigation of ischemia/reperfusion (I-R) injury of water extract of propolis (WEP) by studying the effect of WEP on mesenteric microcirculation of rats subjected to small intestinal I-R. Methods Forty male Wistar rats were randomly divided into sham, I-R and WEP(50, 100,200 mg/kg)groups. Model of small intestinal I-R injury was made by clamping super mesenteric artery for 45 min followed by 120 min of reperfusion in rats. Mesenteric microcirculation was detected at the end of reperfusion. The degree of small intestine injury was assessed with Nackel's method and intestinal wet/dry ratio. HE staining was used to observe the pathology of guts of rats. Superoxide dismutase(SOD) activity and malondialehyde(MDA) content in small intestine were detected. Results (1) WEP alleviated significantly the pathologic lesion of small intestine and resulted in a significant decrease in Nackel's score and intestinal wet/dry ratio, compared with those in I-R group( P 〈 0.01 ). (2) The disturbance of the blood flow in microvessel induced by I-R was improved significantly by WEP. In addition, WEP alleviated significantly the decrease of microcirculatory blood velocity (P 〈 0.05 vs I-R group) and inhibited the adherence of leukocytes to venule ( P 〈 0.01 vs I-R group) in a dose-dependent manner. WEP grOUpr had more activity of SOD and less content of MDA than I-R group( P 〈 0.05). Conclusion WEP protects small intestine from I-R injury by im- proving intestinal microcirculation
