莪术油对骨肉瘤saos-2细胞IGF-1R,Akt及Bcl-2表达的影响

摘    要:目的:初步研究莪术油对骨肉瘤saos-2细胞的作用,并探讨胰岛素样生长因子-1受体(IGF-1R),蛋白激酶B(Akt)及B细胞淋巴瘤/白血病-2(Bcl-2)的表达情况。方法:以saos-2细胞为研究对象,采用MTT法观察不同质量浓度30,60,120 mg·L-1莪术油作用saos-2细胞24,48,72 h后的增殖影响,流式细胞术观察莪术油(30,60,120 mg·L-1)诱导saos-2细胞的凋亡,RT-PCR检测IGF-1R mRNA的表达,Western blot检测Akt及Bcl-2的蛋白表达变化。结果:与空白组比较,莪术油(30,60,120 mg·L-1)能明显抑制saos-2细胞的增殖,并诱导saos-2细胞凋亡,呈剂量依赖性,明显下调IGF-1R mRNA的表达,明显下调Akt及Bcl-2蛋白的表达(P0.05,P0.01)。结论:莪术油具有抑制saos-2细胞生长和诱导凋亡的作用,其抗肿瘤作用与抑制IGF-1R,Akt及Bcl-2的表达有相关性。

Abstract:Objective: To study the effect of Curcumae Rhizoma oil on human osteosarcoma cell line saos-2, and discuss the change of the expression level of insulin-like growth factor-1 receptor (IGF-1R), protein kinase B (Akt) and B-cell lymphoma/leukemia-2 (Bcl-2). Method: The effects of Curcumae Rhizoma oil on the proliferation of osteosarcoma cell line saos-2 were detected by MTT assay. Cytometry was used to analyze cell apoptosis; expression of IGF-1R was detected by real-time PCR, and the expression of Akt and Bcl-2 was detected by western-blot. Result: Curcumae Rhizoma oil could obviously inhibit the growth of osteosarcoma cells and induce saos-2 cell apoptosis in a dose dependent manner. The expression of IGF-1R, Akt and Bcl-2 was down regulated in the curcumol-treated cells. Conclusion: Curcumae Rhizoma oil can inhibit proliferation and induce apoptosis of saos-2 cells, and its antitumor effect is associated with suppressing the expression of IGF-1R, Akt and Bcl-2.
