脑舒胶囊对血管性痴呆大鼠氧化应激损伤的保护

摘    要:目的:观察脑舒胶囊对血管性痴呆(VD)模型大鼠学习能力的影响以及对海马SOD、MDA、皮层GSH—Px表达的影响。方法:筛选雄性SD大鼠72只,随机分为假手术组、模型组、2g/kg、4g/kg、8g/kg脑舒胶囊组和0.2g/kg脑复康组,每组12只。腹腔注射2.5mg/kg硝普钠加反复夹闭双侧颈总动脉制备VD模型;用药组每天灌胃给药1次,模型组和假手术组灌胃给予等容量生理盐水,连续3周。Morris水迷宫检测大鼠学习记忆功能;化学比色法检测海马组织SOD活性及MDA含量,大脑皮层GSH-Px含量。结果:与假手术组相比,模型组大鼠逃避潜伏期显著增加、跨越平台次数、在原平台象限停留的时间、平台象限游程占总游程百分比显著降低,海马组织SOD活性明显降低,MDA含量明显增加、皮层组织GSH—px含量明显降低(P〈0.01);与模型组相比,0.2g/kg脑复康、2g/kg、4g/kg和8g/kg脑舒胶囊大鼠逃避潜伏期缩短、跨越平台次数、在平台象限停留时间增加、平台象限游程占总游程百分比增加(P〈0.05)、大鼠海马组织SOD活性提高、MDA含量降低、大鼠皮层组织GSH-Px活性提高(P〈0.05,0.01).呈剂量依赖性。结论:脑舒胶囊能够改善VD模型大鼠学习能力,可能与提高SOD、GSH—Px活性,降低MDA表达有关。

Abstract:Obiective: To observe the effects of Naoshu capsule on learning and memory, ability, Hippocam pal SOD and MDA, and cerebral cortical, GSH-Px in vascular dementia rats. Methods: 72 screened male SD rats were randomly divided into sham operation group, model group, and 2 g/kg,4 g/kg, 8 g/kg naoshu capsule group and 0.2 g/kg Naofukang group, n = 12. Intraperitoneal injection of sodium nitroprusside 2.5 mg/kg plus repeated bilateral carotid artery occlusion were performed to prepare VD model and medication groups were administered orally once daily(model and sham operation groups were given an equal volume of saline)for 3 consecutive weeks. The changes of learning and memory ability of rats were observed by the Morris water maze test,the content of SOD, MDA and GSH- Px were detected by chemical colorimetric method. Results: Compared with sham operation group, the escape latency and the content of MDA in hippocampus were increased, the frequence of swim ruing over the platform quadrant, length of staying in platform,percentage of swimming length in platform, the activity of SOD in hippocampus, and content of GSH-Px in cortex were decreased significantly in model rats (P 〈 0.01), Compared with model group, the escape latency and the content of MDA in hippocampus were decreased, the frequence of swimming over the platform quadrant, length of staying in platform, percentage of swimming length in platform, the activity of SOD in hippocampus, and content of GSH-Px in cortex were increased significantly in 2 g/kg,4 g/kg,8 g/kg Naoshu capsule groups and 0.2 g/kg naofukang group (P 〈 0.05, 0.01). Conclusion: Naoshu capsule can improve learning ability of VD rats, which may berelated to the increase of SOD, and GSH-Px activity and the decrease of MDA expression.
