不同脱钙液对骨髓组织切片HE染色效果的对比研究

摘    要:目的:探讨不同脱钙液对骨髓组织切片HE染色质量的影响。方法:选用10%乙二胺四乙酸钠(EDTA)、HA S复合酸脱钙液和10%硝酸3种不同的脱钙液,分成A、B、C 3组用于脱钙,通过记录脱钙时间,观察HE染色效果,评价室温下(20~28℃)不同脱钙液对骨髓组织切片制作的影响。结果:A组脱钙液所制切片染色效果良好,但脱钙速度缓慢,所需时间较长;B组脱钙液脱钙速度快,所制切片染色好;C组脱钙液所制切片的染色强度和对比度均不如前两种脱钙液。结论:在制作骨髓组织切片时,EDTA脱钙液和HA S复合酸脱钙液所制切片均能取得良好的染色效果,但EDTA脱钙液脱钙周期过长;HA S复合酸脱钙液脱钙均匀,速度快,更适合于临床速检工作;硝酸脱钙液组染色效果较差。

Abstract:Objective:Discusses the different decalcify fluid to slice the HE dyeing quality to the myeloid tissue the influence.Methods:Selects 10% ethylene diamine four sodium acetate(EDTA),the HAS duplicate complex acids decalcify fluid and 10% nitric acid 3 kinds of different decalcify fluid,divides into A,B,the C 3 groups to use in the decalcify,through the record decalcify time,observes the HE dyeing effect,the appraisal room temperature next(20℃~28℃) different decalcify fluid to the myeloid tissue slice manufacture influence.Results:A group of decalcify fluid makes the section staining effect to be good,but the decalcify speed was slow,needs the time to be long;B group of decalcify fluid decalcify speed was quick,makes the section staining;C group of decalcify fluid makes the slice the dyeing intensity and the contrast gradient were inferior to the first two kinds of decalcify fluid.Conclusion:When manufacture myeloid tissue slice,the EDTA decalcify fluid and the HAS duplicate complex acids decalcify fluid makes the slice to be able to make the good dyeing progress,but the EDTA decalcify fluid decalcify cycle is excessively long;The HAS duplicate complex acids decalcify fluid decalcify is even,the speed is quick,suits in clinical fast examines the work;The nitric acid decalcify fluid group dyes the effect to be bad.
