摘    要:目的:建立快速测定肺癌患者血浆中依托泊苷浓度的方法,考察其治疗窗。方法:采用高效液相色谱法,色谱柱为Waters Symmetry C18,流动相为甲醇-水(45∶55),流速为1 mL·min-1,检测波长为220 nm,进样量为20 μL,柱温为30 ℃。用世界卫生组织(WHO)评价标准判定疗效及观察不良反应以考察治疗窗。结果:依托泊苷血药浓度在2.59~51.8 μg·mL-1范围内线性关系良好(r=0.999 7);方法回收率为94.5%~96.4%;日内、日间RSD均<5.0%。肺癌患者依托泊苷血药浓度治疗窗应为6.525~9.115 μg·mL-1。结论:本方法可快速、准确测定肺癌患者依托泊苷血药浓度;该药在治疗窗内安全、有效。

Abstract:OBJECTIVE:To establish rapid method for the concentration determination of etoposide in plasma of lung cancer patients, and to investigate its therapeutic window. METHODS:HPIC method was adopted. The determination was performed on Waters SymmetryC18(150 mm×4.6 mm,5 μm) column with mobile phase consisted of methanol-water (45∶55) at flow rate of 1 mL·min-1. The detection wavelength was set at 220 nm and injection volume was 20 μL. The column temperature was 30 ℃. The curative effect and result of adverse drug reaction observe were evaluated by WHO criteria to investigate therapeutic window. RESULTS: The linear range of etoposide was 2.59~51.8 μg·mL-1(r=0.999 7) with recovery of 94.5%~96.4%. The RSDs of intra-day and inter-day were all lower than 5.0%. The therapeutic window of etoposide in lung cancer patients wes 6.525~9.115 μg·mL-1. CONCLUSION: It’s quick and accurate to determine plasma concentration of etoposide in lung cancer patients. Etoposide is safe and effective in the range of therapeutic window.
