弹性蛋白酶抑制剂对动脉损伤后血管平滑肌细胞的作用

目的探讨弹性蛋白酶抑制剂对大鼠颈动脉损伤后内膜增厚的影响。方法球囊导管损伤Wistar大鼠颈总动脉内皮,治疗组每日一次给予弹性蛋白酶抑制剂,模型组给予同等剂最的溶剂,空白对照组注射等最的生理盐水。于损伤10d时测定颈总动脉内膜细胞数及面积,用原位末段缺口标记法检测内膜细胞凋亡指数。结果球囊损伤10d后,模型组颈总动脉内膜面积增厚,平滑肌细胞数增多,凋亡指数下降。治疗组较模型组,内膜面积及平滑肌细胞数减少,凋亡指数大幅上升。结论弹性蛋白酶抑制剂能够有效抑制损伤后平滑肌细胞的增殖,诱导平滑肌细胞的凋亡,从而抑制血管损伤后内膜的增厚。

Effects of Elastase Inhibitor on Vascular Smooth Muscle Cells

Objective To investigate the effects ofelastase inhibitor on proliferation of the inner membrane of the artery.Methods The common carotid artery of male Wistar rats was injured by 2F balloon catheter.The rats were divided into three groups:control,model and ONO-6818 groups.On the day that ONO-6818 and model group was injured and after,the rats in ONO-6818 and model groups received once per day garage tube feedings of either ONO-6818 at a dose of 100 mg/kg or administration vehicle,while the rats in the control group were subjected to an equivalent volume of 0.9% saline.On day 10,measure the area and number of VSMC in the inner membrane of common carotid artery,in situ end labeling technique was used for detection of apoptosis of VSMC.Results In model group,area of inner membrane and number of VSMC was increased,and the Apoptosis index was decreased. In ONO-6818 group,area of inner membrane and number of VSMC was decreased and Apoptosis index was increased significantly compared with the model group.Conclusions ONO-6818 inhibits the proliferation of VSMC in inner membrane and induces its apoptosis,thus inhibits the thickening of inner membrane of artery after balloon injury.
