摘    要:目的建立人血浆米非司酮检测的高效液相色谱方法。方法血浆经乙酸乙酯-正己烷萃取,以ZORBAXEclipseXDB—C18为色谱柱;流动相为乙腈-甲醇-水体系,流速为1.0mL·min-1;检测波长310nm。结果米非司酮浓度在25~2000μg·L-1范围内线性关系良好(r=0.9999);定量下限为25μg·L-1;高中低三个浓度的相对回收率分别为101.27±7.31%、99.62±5.53%和100.43±2.63%;日内RSD分别为7.22%、5.55%和2.62%,日间RSD分别为8.64%、5.99%和3.24%。结论本方法简便、快速、准确可靠,适用于人血浆米非司酮浓度的测定及其药代动力学研究。

Abstract:Objective To develop a high performance liquid chromatography method for the determination of mifepriston in human plasma. Methods The plasma was extracted by ethyl acetate and N-Henaxe. The analytical column was packed with ZORBAX Eclipse XDB-C18. The mobile phase was acetonitrile-methanol-water and the flow rate was 1.0 mL ·min-1. The UV detection wavelength was 310 nm. Results Excellent liner relationship was obtained from the range of 25μg·L-1 to 2000μg·L-1 ( r = O. 999 9), and the limit determination was 25μg·L-1. The relative recoveries were 101.27 ± 7.31% , 99.62 ± 5.53% and 100.43 ± 2.63% respectively at three concentrations. The intra-day RSD were 7.22% , 5.55% and 2.62% and inter-day RSD were 8.64% , 5.99% and 3.24% , respectively. Conclusion The method is simple, rapid and accurate and it can be used to determine mifepriston concentration in human plasma and to study its pharmacokineties.
