北京地区哮喘儿童ORMDL3基因表达水平、生活方式与室内空气质量的相关性研究

摘    要:目的 研究北京地区儿童ORMDL3基因表达水平、生活方式、室内空气质量与哮喘发生的相关性.方法 采用病例-对照方法进行实验设计.使用RT-PCR法检测儿童ORMDL3基因表达水平;通过<国际儿童哮喘及过敏研究(ISSAC)问卷>收集儿童生活方式信息;并根据<室内空气质量标准 (GB/T18883-2002)>中规定的检验方法对室内空气中PM2.5,VOCs,甲醛浓度及空气温湿度进行监测.使用t检验、卡方检验分析病例组与对照组儿童ORMDL3基因表达水平,生活方式与室内空气质量差异;使用Logistic逐步回归来分析ORMDL3基因表达水平,生活方式与室内空气质量对与哮喘之间相关性.结果 ORMDL3基因表达水平在病例组与对照组中有显著差异(P<0.05).在早发型哮喘患儿中,ORMDL3基因表达水平、哮喘家族史与哮喘显著相关(P<0.05).而在迟发型哮喘患儿中,ORMDL3基因表达水平、采暖方式、室内空气湿度与哮喘显著相关(P<0.05).结论 ORMDL3基因表达水平与儿童哮喘的发生密切相关,早发型哮喘患儿与遗传因素有显著关联,而迟发型哮喘患儿中遗传因素与环境因素均与哮喘相关.

Abstract:ObjectiveTo Study whether ORMDL3 expression level, life-style and indoor air quality were associated with childhood asthma in Beijing, China. MethodsWe used case-control design to perform our study. Life-style information was collected through ISAAC (International Study of Asthma and Allergies in Childhood) questionnaire results. RT-PCR was performed to test ORMDL3 expression levels. PM2.5, VOC, formaldehyde and humidity in bedrooms and living rooms were monitored by Indoor Air Quality Standard (GB/T18883-2002). T-test, chi-square and logistic regression with stepwise selection (SLE=0.2, SLS=0.2) analysis were used to test the association between ORMDL3 expression level, life-style, indoor air quality and childhood asthma. ResultsExpression level of ORMDL3 was associated with childhood asthma (P<0.05). ORMDL3 expression level and parent asthma history were associated with early-onset childhood asthma (P<0.05), while ORMDL3 expression level, the way of heating and humidity were associated with late-onset childhood asthma. ConclusionORMDL3 expression level was associated with childhood asthma in Beijing, China. Genetic factors were associated with early-onset childhood asthma, and both genetic factors and environmental factors were associated with late-onset asthma.
