摘    要:目的研究新型镇静催眠化合物B2对小鼠脑内单胺类神经递质水平的影响。方法小鼠随机分为溶剂对照组和B2给药组(5mg/kg,ip),采用高效液相色谱-电化学法(HPLC-ECD)检测B2对小鼠不同脑区中去甲肾上腺素(NE)、多巴胺(DA)和5-羟色胺(5-HT)等单胺类神经递质水平的影响;并用比色法测定化合物B2对小鼠下丘脑、纹状体及大脑皮质中单胺氧化酶(MAO)活性的影响。结果与对照组相比,B2(5mg/kg,ip)可使小鼠下丘脑中NE含量由(1.08±0.09)μg/g显著降低至(0.88±0.07)μg/g(P〈0.01);纹状体中DA含量由(12.85±2.21)μg/g显著降低至(8.44±2.25)μg/g(P〈0.01);下丘脑中5-HT含量由(1.12±0.12)μg/g显著降低至(0.56±0.18)μg/g(P〈0.01)。B2对小鼠不同脑区MAO的活性无显著影响。结论B2引起小鼠不同脑区中单胺类递质NE、DA和5-HT水平的下降可能是其发挥镇静催眠作用的机制之一。

Abstract:Objective To study the effect of sedative and hypnotic compound B2 on levels of monoamine neurotransmitters, including norepinephrine (NE) , dopamine (DA) , and 5-hydroxytryptamine (5-HT) in mouse brain. Methods Mice were randomly divided into vehicle group and B2 group (5 mg/kg, ip), then the contents of NE, DA and 5-HT were detected by high performance liquid chromatography-electrochemical detection (HPLC-ECD). The effects of compound B2 on monoamine oxidase (MAO) activities in mouse hypothalamus, striatum and cortex were detected by colorimetrie method. Results B2 (5 mg/kg, ip) could significantly re- duce NE contoent in the hypothalamus from ( 1.08 ± 0. 09 ) μg/g to (0. 88 ± 0. 07 )μg/g ( P 〈 0.01 ) and reduce DA content in the striatum from ( 12. 85 ± 2. 21 ) μg/g to ( 8.44 ± 2. 25 ) μg/g ( P 〈 0. 01 ). 5-HT content in hypothalamus were significantly decreased from ( 1.12 ±0. 12)μg/g to (0. 56 ±0. 18) μg/g (P 〈0. 01 ). There was no significant difference between two groups in MAO activi- ty. Conclusion The sedative and hypnotic effects of B2 might be mediated by the decrease of monoamine levels in different mouse brain regions.
