摘    要:目的对从红豆杉树皮中分离、选育到的1株高产紫杉醇菌种发酵生产新型单体化合物维泰醇(alternol)的发酵工艺进行研究。方法用摇瓶发酵对培养基成分进行单因素、正交设计实验,筛选出合理的基础培养基配方;用5 L罐发酵确定最佳发酵pH值、通气比等;用50 L罐发酵确定放大工艺。结果确定维泰醇的发酵工艺为碳和氮的质量比为40∶80、pH值在6.2~7.2之间、发酵液体积与单位时间内通入空气的体积比(通气比)为1.0∶1.2、溶解氧质量分数(DO)值不得低于起始溶解氧质量分数的10%、比生长速率为10%。50 L罐发酵产量可达420 mg.L-1。结论控制比生长速率10%是工艺放大过程中的关键控制参数。

Abstract:Objective To study the fermentation process of alternol, produced by a paclitaxel producing strain isolated from the bark of taxus yunnanensis. Methods The preferential medium in the flask culture was obtained by single parameter and orthogonal experiment design. Then the suitable pH value and the ratio of air in 5 liters tanks were gained. Finally, the pilot scale process in 50 liters tanks was determined. Results Our results show that the ratio of m(C)∶m(N) was 40∶80, the pH value was 6.2 to 7.2, the ratio of air was 1.0 to 1.2, the DO must be higher than 10 %, the specific growth rate must be maintained at the 10 %.The expression rate of alternol was 420mg/L in the 50 liters tank. Conclusion As the key parameter, the specific growth rate must be controlled at 10 % during fermentation.
