X染色体STR基因座在同胞亲缘鉴定中的应用

目的：探讨X染色体STR基因座独特的遗传方式在同胞亲缘关系鉴定中的应用。方法：应用聚合酶链式反应（PCR）、聚丙烯酰胺凝胶电泳及银染显带技术对1名未知女性个体与3名已知同胞兄妹的22个基因座进行分型，并对线粒体D环高变区作序列测定。结果：得到具有排除价值的2个X染色体基因座（DXS7132、DXS6804）和4个常染色体基因座（D1S549、D2S441、D16S539、D5S818），否定了未知个体与3名同父同母兄妹的亲缘关系。结论：X染色体基因座在男性的基因组中呈单倍型形式存在，特别适用于法医学实践中在父母缺如情况下姐妹之间的亲缘鉴定。

Application of X-chromsone SIR loci in the siblings' identification

Objective：To study the in siblings＇ identification. Methods application of specific genetic pattern of X-chromosome STR loci 22 STR loci of whole blood of suspicious one and three siblings were examined with polymerase chain reaction （PCR）, PAGE and silver staining technique, and the hypervariable region of the mitochondrial D-loop were sequenced. Results： Two X-chromosome STR loci （DXS7132. DXS6804） and Four autosomal STR loci （D1S549, D2S441, 4D16S539 and D5S818） have been found, which can be used to exclude the suspicion in siblings＇ identification. Conclusion： X-chromosome STR loci exist in the male genome in the form of haploid, which is especially useful for forensic science application, such as the siblings＇ identification in the absence of parents.
