大孔吸附树脂纯化桔梗总皂苷的工艺研究

摘    要:目的运用大孔吸附树脂分离纯化桔梗总皂苷,为桔梗总皂苷的生产提供参考。方法采用单因素试验结合正交试验法,以桔梗总皂苷含量、保留率为指标,对桔梗总皂苷的纯化工艺进行优选。结果最佳条件为选用D101型大孔树脂,上样浓度0.5g生药材/ml,径高比1:9,吸附流速为2BV/h,吸附2h后,以6BV水洗脱,再分别以5BV25%、70%乙醇洗脱,解析流速为3BV/h,收集70%乙醇洗脱液,纯化后桔梗总皂苷的含量达到56.32%,保留率为74.86%。结论 D101树脂对桔梗总皂苷具有较好的吸附和解析性能,纯化工艺简单可行、重复性好,具有良好的产业化前景。
