蒙药顺气补心十一味丸对心肌梗死大鼠心肌血管生成和VEGF表达的影响

摘    要:目的探讨顺气补心十一味丸对心肌梗死后缺血心肌血管生成的作用以及可能机制。方法健康成年Wistar大鼠72只,随机分为模型对照组、蒙药低剂量组、蒙药高剂量组、卡托普利组,每组18只。所有大鼠背部皮下多点注射盐酸异丙肾上腺素(ISO)复制心肌梗死模型,模型建立后,药物干预组分别给予顺气补心十一味丸0.461、1.844 g/kg和卡托普利25 mg/kg,每日两次灌胃。分别取6只大鼠在7、28 d和42 d处死,免疫组化染色观察CD34和血管内皮生长因子(VEGF)蛋白表达。结果①MVD结果显示:7、28 d和42 d蒙药低、高剂量组、卡托普利组与模型对照组相比MVD明显增加(P<0.01);②VEGF蛋白在7、28 d和42 d时蒙药低、高剂量组与模型对照组相比表达明显增强(P<0.01);卡托普利组与模型对照组相比无差异。③病理结果显示:相应时间点各治疗组较模型对照组坏死灶面积缩小,42 d时心肌纤维化程度减轻。结论顺气补心十一味丸能够促进心肌梗死大鼠缺血心肌的血管生成,能上调缺血心肌的VEGF蛋白表达,其促血管生成的机制可能与上调缺血心肌的VEGF蛋白表达有关。

Abstract:Objective To observe the role and mechanisms of Mongolian drug Shunqi Buxin Shiyiwei pill on angiogenesis in ischemic myocardium rats.Methods 72 Wistar adult rats were randomly divided into model,Mongolian drug low-,high-dose and captopril groups.Myocardial infarction models were established by injecting isoproterenol hydrochloride(ISO).Rats in intervention groups were given Shunqi Buxin Shiyiwei pill 0.461,1.844 g/kg,captopril 25 mg/kg twice a day until the end of the experiment.6 rats were killed on 7,28 and 42 days respectively.The expressions of CD34 and VEGF were detected by immunohistochemical staining.Results MVD was increased in Mongolian drug low-,high-dose and captopril groups compared to model group(P<0.01) at 7,28 days and 42 days.VEGF expression was enhanced in Mongolian drug low-and high-doses group compared to model group(P<0.01).There was no significant difference between captopril and model groups.Area of myocardium infarction was decreased in each intervention group.Conclusions Shunqi Buxin Shiyiwei pill can promote angiogenesis of myocardial infarction boundary by up-regulating VEGF expression.
