健康人口服盐酸黄连素片剂后的尿药分析与药物代谢初步研究

摘    要:本文以13-甲基小檗碱为内标,用正相高效液相色谱-荧光检测的方法定量分析尿液中的小檗碱.考察了健康人一次口服 0.2g 盐酸黄连素糖衣片后 96h内小檗碱尿药排泄量变化,按矩量法求得其体内药物平均滞留时间(MRT)为32.63±6.15 h,累积尿药排泄量为 27.49±4.74μg;首次检测到小檗碱在人体内有四个未知的代谢峰.

Abstract:A normal phase liquid chromatographicmethod (flurorescent detector) for determinating berberine (BER) concentration in humanurine was reported. The 13-methylberberine was used as the internal standard. A mixture ofdichloromethane-methanol-diethyl amine-acetic acid ( 90 90.40.5) was served as themobile phase. The dynamic change of excretion of BER in urine after oral administration of0.2 g BER.HCL tablets in single dosage was observed and MRT and AUC was calculed to be32.63±6.15 h and 27.49±4.74μg by statistical moment theory. Furthermore there were four metabolites ofberberine chloride in human urine were detected for the first time.
