非酒精性脂肪性肝病大鼠肝脏SSeCKS的表达

目的:探讨Src抑制的蛋白激酶C底物(SSeCKS)在大鼠非酒精性脂肪性肝(NAFL)形成中的作用。方法:SD大鼠16只,随机分为正常对照组(正常组)和高脂饮食组(高脂组)。高脂饲料喂养14周后,观察肝脏组织病理改变;检测大鼠血脂及肝功能;免疫组化和westernblot方法研究大鼠脂肪肝形成中SSeCKS表达变化。结果:高脂组大鼠肝脏指数显著高于正常组(P均<0.01)。高脂组总胆固醇(TC)、甘油三酯(TG)、低密度脂蛋白-胆固醇(LDL-C)及丙氨酸转氨酶(ALT)均显著高于正常组(P均<0.01)。肝组织SSeCKS蛋白表达亦明显增强。结论:非酒精性脂肪肝大鼠肝组织SSeCKS表达变化与脂肪肝引起的脂质过氧化损伤程度密切相关。

Expression of SSeCKS in nonalcoholic fatty liver of rats

Objective:To investigate the role of Src suppressed C kinase substrates (SSeCKS) in the nonalcoholic fatty liver (NAFL) of rats. Methods:Sixteen SD rats were randomly divided into two groups: the control group and high-fat diet induced fatty liver group. The expression of SSeCKS in the hepatocytes was examined by immunohistochemistry and western blot. Meanwhile, the levels of total cholesterol (TC), serum triglyceride (TG),low-density lipoprotein cholesterol(LDL-C), and alanine aminotransferase (ALT) were measured by biochemical method. Results:The expression of SSeCKS and the number of SSeCKS positive cells were markedly increased, and the levels of serum TC, TG, ALT in the fatty liver rats were increased as compared with those in the normal controls. Conclusion:The increase of SSeCKS expression in NFAL is closely associated with the severity of liver inflammation and damages. SSeCKS may play an important role in the progression of rat NAFL.
