降钙素基因相关肽对局灶性脑缺血再灌注大鼠脑组织P53蛋白表达的调节作用

目的　研究降钙素基因相关肽(CGRP)对局灶性脑缺血再灌注大鼠脑组织P53蛋白表达的影 响,探讨降钙素基因相关肽对脑组织缺血再灌注损伤的保护作用及机制。方法　用线栓法制备大鼠大脑中动 脉阻塞(MCAO)模型,应用免疫组化和显微图像分析方法检测大鼠脑缺血再灌注后脑组织P53蛋白的表达。 结果　假手术组海马和顶叶内未见P53阳性细胞,脑缺血再灌注组阳性细胞明显增多,注射CGRP后P53阳 性细胞平均灰度值明显高于缺血再灌注组(P<0.01)。结论　CGRP下调缺血神经元P53蛋白的表达,可能 对缺血神经元的恢复有促进作用。

The Effects of Calcitonin Gene Related Peptide on the Expression of P53 Protein during Focal Cerebral Ischemia/Reperfusion in Rats

Objective To study the effects of calcitonin gene related peptide(CGRP) on the expression of P53 protein in brain tissue of rats with focal cerebral ischemia/reperfusion(I/R),and to explore the protective effects and mechanism of CGRP on ischemia brain tissue. Methods Using middle cerebral artery occlusion (MCAO) to make focal cerebral ischemia model, the expression of P53 protein was detected with immunohistochemical method and analyzed by microimage analysis system. Results P53 protein positive cells in hippocampus and parietal lobe of rats were not found in sham group, but were found a lot in focal cerebral I/R group.The average gray values of P53 protein positive cells in hippocampus and parietal lobe were higher in CGRP group than in I/R group(P<0.01). Conclusion Downregulation of CGRP on the expression of P53 protein in ischemic neurons might promote the recovery of the ischemia neurons.
