盐源山蛭注射液治疗脑出血的实验研究

摘    要:目的 :探索纯化的盐源山蛭注射液促进血肿吸收的作用及其作用机理。方法 :采用定量胶源酶注入大鼠尾状核建立脑出血模型 ,观察盐源山蛭注射液对鼠脑内血肿容积、脑水含量、血浆 F。 a活性、 AT- 活性、 D- D含量及组织病理变化的影响。 BWC采用称重法 ,酶活性测定采用发色底物法 ,D- D含量测定采用 EL IAS测定。结果 :(1)盐源山蛭注射液治疗后 6 d、 10 d的脑内血肿明显缩小 ;使治疗后 3d、 6 d的 BWC明显减轻。 (2 )盐源山蛭注射液能抑制脑出血后 F。 a活性的升高 ,不影响 AT- 活性 ,增加纤溶活性。 (3)盐源山蛭注射液能加快脑出血后的病理组织修复。结论 :(1)盐源山蛭注射对大鼠脑出血后脑内血肿有治疗作用 ;(2 )作用机制可能为 :拮抗凝血形成的稳定因素 ,激活内源性纤溶系统 ,促进胶质细胞增生 ,改善微循环

Abstract:Objective: to investigate the effect of purified haemadipsa yanyuanensis injection on intracerebral hematoma in rats and its potential mechanism. Methods: We established the experimental ICH model in rats by stereotaxically injecting quantitative collagenase into their left caudate nucleuses. The effects of haemadipsa yanyuannesis injection on hematoma volume, brain water content(BWC), thromabin(FⅡa)activity, antithrombin Ⅲ(AT-Ⅲ)activity and D-dimer (D-D) content in plasm, as well as histo-pathological changes were observed. BWC was calculated by drying-weighing method. Activities of F.Ⅱa and AT-Ⅲ were measured by colorimetric assay. D-D content was determined by ELISA. Results: (1)Haemadipsa yanyuanensis injection can quicken intracerebral hematoma in rats to liquidize and absorb, significantly decreasing the hematoma volume on the 6th and 10th day ( P<0.05 ), as well as reduce bran edema, markedly decreasing BWC both at 72 hours and on the 6th day( P<0.05 ). (2)Haemadipsa yanyuanensis injection can inhibit the ascendance of thrombin activity, have no effect on antithrombin-Ⅲ activity, and increase fibrinolysis activity in plasm in rats after ICH, which would advantage the absorption of liquified blood. (3)Haemadipsa yanyuanensis injection can improve histo-pathologic changes. Conclusion: (1)Haemadipsa yanyuanemsis injection has a treatment effect on intracerebral hematoma in rats after ICH. (2)The function mechanisms of haemadipsa yanyuanensis injection on cerebral hematoma in rats after ICH may be associated with the disruption of the stability of hematoma formation, activation of endogenous fibrinolytic system, improvement of microcirculation and acceleration of glial cells hyperplasia.
