表没食子儿茶素没食子酸酯对阿米卡星诱导大鼠螺旋神经元Bcl-2和Bax表达的影响

摘    要:目的 探讨表没食子儿茶素没食子酸酯[(-)-epigallocatechin-3gallate,EGCG]对阿米卡星(amikacin,Amk)诱导的大鼠耳蜗螺旋神经元(spiral ganglion neuron,SGN)Bcl-2和Bax表达的影响.方法 设生理盐水对照组、Amk[450 mg/(kg·d)×14 d]损伤组和Amk加EGCG[50 mg/(kg·d)×14 d]保护组.各组动物均深度麻醉后半身灌注固定处死,取右侧耳蜗标本固定、脱钙、石蜡轴住包埋、轴位切片,近中轴位切片行Bcl-2和Bax免疫组化染色,光学显微镜下检测耳蜗SGN表达情况、并通过切片图像分析进行半定量处理比较.结果 对照组SGN Bcl-2染色胞浆有棕黄色或黄色颗粒或斑片呈中度着色、Bax染色胞浆有棕黄色或黄色颗粒或斑片呈轻度着色[SGN Bcl-2和Bax染色平均灰度值分别为(81.24±3.10)和(90.92±3.62)1,损伤组SGN Bcl-2染色胞浆着色减轻、Bax染色胞浆着色加深fSGN Bcl一2和Bax染色平均灰度值分别为(99.58±3.97)和(71.12±2.98)1,保护组SGN Bcl-2染色胞浆着色加深、Bax染色胞浆着色减轻[SGN Bcl-2和Bax染色平均灰度值分别为(82.86±8.10)和(89.15±2.85)],图像分析结果 显示损伤组与对照组、损伤组与保护组之间的SGN Bcl-2和Bax染色平均灰度值差异有显著性.结论 Amk耳中毒诱导耳蜗螺旋神经元氧化损伤时Bcl-2表达下调,Bax表达增强.EGCG可以减弱Amk耳中毒时耳蜗螺旋神经元Bax的表达,同时增强Bcl-2的表达,从而减轻Amk耳毒性.

Abstract:[Objective] To study the expression of Bcl-2/Bax in Amikacin (Amk)-induced spiral ganglion neuron(SGN) apoptosis in the rat cochlea and the protection of (-)-epigallocatechin-3-gallate(EGCG).
