血管紧张素Ⅱ对培养乳鼠非心肌细胞DNA及蛋白合成的影响

摘    要:AugⅡ作为一种独立于血液动力学及神经体液因子,对哺乳动物心脏细胞的直接作用尚未完全清楚。本实验中,我们观察了AugⅡ对培养心脏的非心肌细胞的作用,结果表明:在培养的非心肌细胞,AugⅡ引起蛋白合成、DNA合成及细胞数目的明显增加,但不影响细胞的体积。AugⅡ在非心肌细胞诱导的DNA合成受AugⅡ受体拮抗剂[Sar1,Val5,Ala8]-AugⅡ完全抑制、这些结果提示:(1)AugⅡ诱导心脏非心肌细胞的增殖;(2)AngⅡ的上述作用通过AT1受体亚型调控。

Abstract:Increasing evidence suggests that angiotensin Ⅱ(AugⅡ)may act as a growth factor for the heart.However,direct effects of Aug Ⅱon cardiac nonmyocytes-independent of secondary hemodynamic and neurohumoral effects have not been well characterized.In this experiment, we observed effects of Aug Ⅱon cultured cardiac nonmyocytes.The results showed that Ang Ⅱ1,10,100,1000nmol·L-1 induced increases in protein synthesis,DNA synthesis and cell number without changing cell volume in nonmyocytes.The Aug Ⅱ-induced DNA synthesis in nonmyocytes was fully blocked by[Sar1,Val5,Ala8]-Aug Ⅱ which suggested that:(1) Aug Ⅱcaused hyperplasla of cardiac nonmyocytes(2)The effects of AugⅡ examined were mediated primarily by the AT1 receptor subtype.
