| 摘 要： | 目的 研究穿琥宁对人大肠癌HCT-8细胞生长的影响。方法 通过生长曲线测定、MTT、流式细胞仪、电镜等检测、分析方法，探讨穿琥宁对HCT-8的抑制增殖及诱导凋亡作用。结果 生长曲线、MTT表明不同浓度穿琥宁对HCT-8细胞有不同程度的抑制作用。流式细胞仪检测穿琥宁有一定的致凋亡作用。电镜下，细胞呈圆形改变，胞膜外微绒毛明显增多，核浆比例减少。结论 穿琥宁对HCT-8细胞增殖有一定抑制作用，并诱导其成熟分化、凋亡。

| Abstract: | Objective To study the effect of Chuanhuning on HCT-8 cells' proliferation. Methods Growth curve was evaluated by MTT assay, apoptosis was detected by morphological assay and flow cytometry(FCM), and apoptotic cells were observed electromicroscopically after HCT-8 cells were cultured by Chuanhuning. Results Chuanhuning showed dose-dependent and time-dependent inhibitory effect on the proliferation of HCT-8 cells. Differentiation and apoptosis were observed under electronic microscope. Conclusion Chuanhuning can induce apoptosis and differentiation of HCT-8 cells of human colorectal carcinoma, and inhibit proliferation of HCT-8 cells. |
