解毒活血汤联合微囊化肝细胞移植对急性肝衰竭大鼠Caspase-8、BID的影响

摘    要:目的解毒活血汤联合微囊化肝细胞移植对急性肝衰竭大鼠Caspase-8、Bid的影响。方法将急性肝衰动物模型随机分成模型组、裸肝细胞移植组、微囊化肝细胞移植组、解毒活血汤联合微囊化肝细胞移植组。造模后24 h、48 h、72 h、120 h和168 h测定ALT、AST,免疫组化法检测Caspase-8、Bid表达。结果解毒活血汤联合微囊化肝细胞移植组ALT、AST下降,48 h、72 h与裸肝组、微囊组比较差异有统计学意义(P<0.05);解毒活血汤联合微囊化肝细胞移植组Caspase-8、Bid表达下降,48~168 h较裸肝组、微囊组有统计学意义(P<0.05)。结论解毒活血汤联合微囊化肝细胞移植显著改善肝衰大鼠肝功能及预后。

Abstract:Objective To observe the development of Caspase-8 and Bid in acute liver failure's rat treated by microen- eapsulation hepatocytes transplantation and Jieduhuoxue soup. Methods The rats of acute liver failure induced by D- galactosamine were individed four groups: groupl was model group,group 2 was treated by non-microen-capsulated hepatocytes transplantation, group 3 was treated by microencapsulated hepatocytes transplantation,group 4 was treated by mi- croencapsulated hepatocytes transplantation and Jieduhuoxue soup. Testing four groups rats' hepatic biochemistry and the protein of Caspase-8 and Bid by immunohistochemfstry at 24,48,72,120 and 168 hours. Results Alanine aminotransferase (ALT) and aspartate aminotransferase(AST) in group 4 were decreased Obviously. ALT and AST in group 4 had statistical significance with group 2 and group 3 at 48 and 72 hours(P 〈 0.05). The protein of Caspase-8 and Bid in group 4 were decreased obviously. It had statistical significance with group 2 and group 3 at 48,72,120 and 168 hours (P 〈 0.05 ). Conclusion Microencapsulated hepatocytes transplantation and Jieduhuoxue soup can improve rat's liver biochemistry and the prognosis of acute liver failure's rat.
