腺苷对冠心病患者白细胞CD11b/CD18表达的影响

摘    要:目的探讨不同剂量腺苷对不同类型冠心病患者单核细胞及中性粒细胞表面CD11b/CD18表达的影响。方法选择经临床症状、心电图改变及心肌酶学确诊的冠心病患者及正常健康者采血,患者血液分别给予不同剂量的腺苷进行干预,免疫荧光标记后用流式细胞仪分别检测正常人及患者用药前后单核细胞与中性粒细胞CD11b/CD18的表达。结果冠心病患者中性粒细胞与单核细胞CD11b/CD18的表达较正常人明显升高(P<0.01),而不稳定型心绞痛与急性心肌梗死患者其CD11b/CD18表达差异无统计学意义(P>0.05);腺苷浓度在10、30、100μmol/L能有效降低冠心病患者单核细胞及中性粒细胞CD11b/CD18的表达(P<0.05),但30、100μmol/L组之间差异无统计学意义(P>0.05)。结论冠心病的发生可能与CD11b/CD18表达升高有关,腺苷可通过降低CD11b/CD18的表达而降低白细胞与内皮细胞之间的黏附性。

Abstract:Objective To explore the effect of different doses of adenosine on the expression of adhesion molecules CD11b/CD18 in neutrophils and monocytes in patients with coronary heart disease(CHD).Methods The patients(who were diagnosed by clinical symptoms,changes of electrocardiogram and myocardial biochemical markers)and normal healthy individuals were drew blood,the blood of patients was given different doses of adenosine to intervene.The expression of CD11b/CD18 in neutrophils and monocytes of normal people and patients were measured by direct immunofluorescence techniques associate with flow cytometry.Results Expression of CD11b/CD18 in neutrophils and monocytes in CHD patients were markedly higher than those in normal cases(P<0.01),but expression of CD11b/CD18 in acute myocardial infarction(AMI)and unstable angina pectoris(UAP)were not significantly different(P>0.05).Adenosine can reduced expression of CD11b/CD18 in neutrophils and monocytes in 10,30,100 μmol/L(P<0.05),but expression of CD11b/CD18 in 30 μmol/L and 100 μmol/L were not significantly different(P>0.05).Conclusion The incidence of coronary heart disease may be related with the increase of CD11b/CD18 expression.Adenosine may reduce adhesion between white blood cell and endothelial cell by reducing CD11b/CD18 expression.
