摘    要:目的研究CD1小鼠空肠cajal间质细胞(ICC)的分离和培养方法,为进一步研究其生理学特征提供基础。方法无菌条件下取小鼠空肠组织,分离出平滑肌肌条,采取组织块培养法,在24孔培养板中进行原代培养,光镜下观察其形态,用荧光标记的特异性C_Kit抗体进行鉴定。结果倒置显微镜下观察ICC有多个突起,并有次级分支,相互之间形成网络。免疫荧光法可见ICC呈阳性。结论组织块培养法可以简便、有效地培养ICC。所培养的ICC细胞生物学特性有待进一步研究。

Abstract:Objective To study the method for islation and culture of ICC (interstitial cells of Cajal) in jejunum of CD1 mice and offer the base for investigating the biologic property of ICC.Methods The jejunum segments of mice were dissected in asepsis and the smooth trips were isolated,the primary culture was used in 24_well culture dishes by primary histoculture.the cell appearance was obserued in light microscope and identified with fluorescently_labeled specific C_Kit antiboby.Results There were more than one primary processes that branched into secondary processes in ICC cells and the network was formed between them. The culture ICC was positive by fluorescent staining with C_Kit antibody.Conclusion The method for culturing ICC is simple and efficient,and its biological property is needed to be studied.
