摘    要:目的:以石蜡包埋组织为材料进行DNA甲基化特异性PCR(MSP)检测,研究其可行性。方法:40例食管组织以中性福尔马林固定、包埋、脱蜡、DNA样品制备和亚硫酸盐修饰后进行PCR测定。2例MSP阳性的冰冻甲状腺组织以70%酒精、丙酮和中性福尔马林分别固定及石蜡包埋后,非甲基化引物PCR扩增,比较不同固定剂的MSP效果。结果l石蜡包埋食管组织MSP检出率与文献报道的冰冻组织MSP检出率相同,三种常用方法固定的甲状腺组织均能获得理想的MSP效果。结论:MSP通过选择合适的实验条件,可应用于石蜡包埋组织。

Abstract:Objective To test the methylation-specific polymerase chain reaction (MSP) on the parafin-embedded tissues. Methods Forty esophageal samples were fixed by neutral buffered formalin, then parafin-embedded and detected by MSP; 2 MSP positive thyroid samples were cut into 3 parts and fixed by 70% ethenal, neutral buffered formalin and acetone, then parafin-embedded and detected by MSP. Results There was no significant difference between the parafin-embedded esophageal tissues and the frozen tissues (in reference) on MSP positivity; All three fixation methods had fine outcomes on MSP positive thyroid tissue. Conclusion The MSP methods can be successfully used on parafin-embedded tissues.
