摘    要:目的:探讨普洱茶水提取物对丝裂霉素C(mitomycin C,MMC)诱发的人成淋巴细胞株遗传损伤的影响。方法:以正常人成淋巴细胞株GM12593为实验材料,设计对照组、MMC作用组和普洱茶组不同条件培养细胞株。对照组用RPMI-1640正常培养,MMC作用组培养基中MMC的终浓度为0.1μg/ml,普洱茶组培养基中除含有0.1μg/ml的MMC外,还分别加入浓度为0.125、0.25、0.5和1 mg/ml的普洱熟茶和生茶水提取物。培养24 h后加入细胞松弛素B,作用28 h后制片,计数不同培养条件下的双核细胞微核率。结果:普洱熟茶在0.125和0.25 mg/ml,普洱生茶在0.125 mg/ml时,均明显降低由MMC引起的细胞微核率(P〈0.05);普洱熟茶降低MMC诱发的细胞微核率的作用略优于普洱生茶,但两者间的差异无统计学意义。结论:一定浓度的普洱茶能降低由MMC诱发的人成淋巴细胞株的遗传损伤。

Abstract:OBJECTIVE:The study focused on the effects of the water extract of Pu’er tea on genetic damage induced by mitomycin C(MMC) in human lymphocyte cell line.METHODS:The lymphoblast cell line GM12593 was cultured in RPMI-1640 medium plus MMC(0.1μg/ml) and different combinations of red and raw Pu’er tea(0~1 mg/ml) for 24 h.Cytochalasin B was added and cells were cultured for another 28 h,centrifuged and put on slides.The frequencies of micronucleated binucleated cells were analysed.RESULTS:Fermented(red) Pu’er tea(0.125 mg/ml,0.25 mg/ml) and raw(green) Pu’er tea(0.125 mg/ml) significantly reduced the frequency of micronucleated binucleated cells induced by MMC(P<0.05).The effect of fermented Pu’er tea in preventing genetic materials induced by MMC was slightly stronger. CONCLUSION:Pu’er tea was able to reduce the genetic damage induced by MMC.
