脂联素与非酒精性脂肪性肝病相关性分析

摘    要:目的:建立高脂饮食诱导的NAFLD大鼠模型,初步探讨脂联素在NAFLD发病机制中的作用以及其与肝脏脂肪变性和炎症程度的相关性。方法:将Wistar雄性大鼠随机分成2组,每组15只。正常对照组喂养普通饲料12周、模型组予高脂饲料喂养12周。12周后处死全部大鼠,称体重和肝湿重,计算肝指数,行肝脏的病理检查,判断肝脏脂肪变性程度、计算肝脏炎症积分,生化法测定血清谷丙转氨酶(ALT)、血清谷草转氨酶(AST)、血清甘油三酯(1、G)、总胆固醇(TC)、肝匀浆TG、TC,酶联免疫吸附试验法(ELISA)检测血清脂联素。结果:高脂饮食12周后成功建立了NAFLD大鼠模型。与对照组比较,模型组大鼠肝细胞呈中一重度脂肪变性,肝脏炎症积分显著升高(P〈0.01);肝湿重及肝指数明显增高(P〈0.01);血清ALT(P〈0.05)、AST、TC值以及肝匀浆TG、TC值明显升高(P〈0.01);血清脂联素值明显降低(P〈0.05);血清脂联素水平与肝脏脂肪变性及炎症积分无相关性,与肝匀浆TC呈负相关(r=-0.638,P=0.038)。结论:NAFLD存在低脂联素血症,脂联素在NAFLD的发病中可能有一定的作用,但与NAFLD肝脏脂肪变性和炎症积分无相关性。

Study on the Relationship between Adiponectin and Nonalcoholic Fatty Liver Disease

Abstract:Objective: Establish rat model of NAFLD in order to evaluate the role of adiponectin in the pathogenesis of NAFLD, to appraise the relationship between adiponectin and the degree of steatosis and inflammation in the livers of rats with NAFLD. Methods:Thirty male Wistar rats were divided into two groups with fifteen for each randomly. Mod- el group were fed with high fat diet while the normal control group with common diet for 12 weeks. After 12 weeks ex- periment, all rats were sacrificed. Histopathological changes in liver, liver wet weight and liver index were observed. The 1event of AST and ALT in serum, TG, TC in serum and liver tusse homogenates were measured by biochemical methods. Serum adiponectin was detected by ELISPu Results:The NAFLD rat model was successfully established with fat diet for 12 weeks. Compared with the normal control group, liver index, histological activity index, level of serum AST, TC and TC, TG in liver homogenates(P〈0. 01), serum ALT(P〈0. 05)and decrease of serum adiponectin. There was no relationship between serum adiponetin and the degree of steatosis and inflammation in the livers of rats with NAFLD. Condusion: Hypoadiponectinemia was found in patients and rats with NAFLD, adiponectin might play a role in the pathogenesis of NAFLD. There was no relationship between serum adiponetin and the degree of steatosis and inflammation in the livers of rats with NAFLD.
