摘    要:目的观察益气活血方对心肌梗死后心力衰竭大鼠心功能的影响并探讨其中的细胞凋亡机制。方法将50只雄性SD大鼠随机分为假手术组、模型组、酒石酸美托洛尔组、益气活血小剂量组和益气活血大剂量组。所有组别均进行大鼠冠状动脉结扎术,假手术组不进行最后的冠状动脉结扎。益气活血方由人参、黄芪、当归按一定比例组成。酒石酸美托洛尔组按12 mg/(kg·d)给药剂量灌胃,益气活血小剂量组和大剂量组分别按2 g/(kg·d)和20 g/(kg·d)给药剂量灌胃,假手术组和模型组给予生理盐水灌胃。于造模后24小时开始,连续给药8周后进行小动物心动超声检测以及心脏组织取材TUNEL染色检测细胞凋亡。结果小动物心动超声结果显示,与假手术组比,模型组大鼠的心脏LVDd、LVDs显著增加(P<0.05),LVEF%、LVFS%显著降低(P<0.05)。与模型组比,给予益气活血方小剂量可以显著改善LVEF%、LVFS%(P<0.05),而大剂量益气活血方有改善LVEF%、LVFS%的趋势,但差异没有统计学意义(P>0.05)。大鼠心肌组织TUNEL染色表明,模型组大鼠心肌细胞TUNEL染色阳性细胞核显著增高(与假手术组比较,P<0.01),益气活血小剂量和大剂量可以明显抑制冠状动脉结扎大鼠模型心肌组织细胞凋亡的发生(P<0.05;P<0.01)。结论益气活血方可以改善心肌梗死后心功能,抑制细胞凋亡的发生可能是其中重要的机制。

Yiqi Huoxue Recipe on cardiac function in rats with heart failure after myocardial infarction and apoptotic mechanism in it

Abstract:Objective To observe the effect of Yiqi Huoxue Recipe on cardiac function in rats with heart failure after myocardial infarction and to investigate the mechanism of the cell apoptosis. Methods 50 male SD rats were randomly divided into control group, model group, Betaloc group, Yiqi Huoxue low dose group and high dose group. All groups of rat were treated with coronary artery ligation operation, except control group without coronary artery ligation. Yiqihuoxue recipe composed of ginseng, astragalus and Angelica sinensis according to a certain proportion. Rats in Betaloc group were treated with 12 mg/( kg· d) Betaloc by garage, and Yiqi Huoxue low dose and high dose group rats threated respectively with 2 g/ (kg·d) and 20 g/(kg·d) dosage Yiqi Huoxue Recipe by garage. Contrl group and model group were given saline by gavage. All the treatment started after 24 hours after operation and for 8 weeks. Then the animals were detected with small animal heartbeat ultrasonic and the heart tissues were stained by TUNEL for apoptosis. Results Small animal heartbeat ultrasound results showed that model group rats cardiac LVDd, LVDs increased significantly (P 〈 0. 05 ) and LVEF% , LVFS% decreased significantly (P 〈 0. 05 ) compared with that of control group rats. Compared with model group, low dose Yiqi Huoxue Recipe can signif- icantly improve LVEF%, LVFS% ( P 〈 0. 05 ), and high dose of Yiqi Huoxue Recipe can improve LVEF%, LVFS% trend, but the difference was not significant (P 〉 0. 05). Rat myocardial tissue TUNEL staining showed that TUNEL positive myocardial cells of rats in the model group increased significantly compared with sham operation group, (P 〈0. 01 ) , and low dose and high dose Yiqi Huoxue Recipe can inhibit the apoptosis of myocardial cells in rats with coronary artery ligation (P 〈0. 05, P 〈0. 01 ). Conclusion Yiqi Huoxue Recipe composed of ginseng, astragalus and angelica can improve cardiac function after myocardial infarction in rats. Apoptosis inhibition is one of the most important mechanisms.
