低氧诱导心肌细胞表达血管内皮生长因子发生机制的实验研究

摘    要:目的 :研究低氧诱导心肌细胞表达分泌血管内皮生长因子(VEGF)的发生机制。方法 :将新生SD大鼠的心肌细胞进行原代培养 ,培养的细胞分为二组 :正常对照组和低氧培养组 ,低氧组采用向培养瓶内灌注 99.99%N2 的方法 ,2 4h后将预置的盖玻片取出 ,进行免疫组织化学检测分裂原活化蛋白激酶 (MAPK)活性。结果 :低氧培养的心肌细胞MAPK活性高于对照组 (P <0 .0 0 1)。结论 :MAPK通路可能是低氧促进心肌细胞表达和分泌VEGF蛋白的机制之一

Abstract:Objective: To study experimental studies on the mechanism of development for the expression of cardiomyocytes on VEGF by induced hypoxia in rat.Method: The neonatal rat cardiomyocytes were primary cultured. These cardiomyocytes were divided into 2 groups: normal culture group and hypoxia-induced group. The hypoxic group was done by irrigating 99.99% N 2. After 24 hours, the cover glasses being lay up in advance were gained. The MAPK expression was measured by immunohistochemistry.Results: MAPK expression increased significantly in cultured cardiomyocytes exposed hypoxia.Conclusion: MAPK pathway is one of the mechanism of VEGF expression and secretion in cardiomyocytes.
