摘    要:目的探讨雷公藤内酯醇(Tri)对Wistar大鼠实验性自身免疫性脑脊髓炎(EAE)的可能治疗机制。方法将40只EAE大鼠动物随机分为模型组及高低剂量的Tri治疗组,并以氢化可的松(HC)作为阳性对照,每组各10只,再取10只注射生理盐水为空白对照,观察Tri对EAE动物模型作用程度、脑和脊髓的病灶数目及血清肿瘤坏死因子-α(TNF-α)、IL-10的影响。结果Tri减轻EAE动物发病程度、减少脑和脊髓的病灶数目,降低血清TNF-α水平的同时升高IL-10的水平。结论Tri治疗EAE的作用具有一定效量关系,其机制可能与抑制TNF—α并上调IL-10的免疫应答有关。

The effect of triptolide on experimental autoimmune encephalomyelitis in rats and its mechanism

Abstract:Objective To investigate the therapeutic efficacy and mechanism of Triptolide (Tri) on the treatment of experimental autoimmune encephalomyelitis (EAE) in rats. Methods Tri was administered with two different doses in Wister rats, and hydrocortisone ( HC) was used as control. The severity of the disease was scored on a scale 0-3 according to symptoms. Histological examinations were performed in the sections of brain and spinal cord sections. The serum TNF-α and IL-10 were examined by ELISA. Results Tri treatment with two different doses had a significantly protective action on the disease model. The protective role of Tri was related with significant reduction in the numbers of inflammatory focus in the brain and spinal cord and decrease of the levels of serum TNF-α and IL-10.Conclusion The therapeutic efficacy of Tri have a dose-dependent manor, the mechanism of Tri on EAE may be related to down-regulate the expression of TNF-α and up-regulate the expression of IL-10.
