粒细胞-巨噬细胞集落刺激因子对前列腺癌细胞PC-3M端粒酶活性的影响

摘    要:目的 研究粒细胞-巨噬细胞集落刺激因子(GM-CSF)对前列腺癌PC-3M细胞株端粒酶活性的影响。方法 以不同浓度的GM-CSF作用于PC-3M细胞,采用Telomerase PCR ELISA方法定性和定量检测端粒酶活性改变。结果 与空白组相比,10、20、40和100ng/m1的GM-CSF能使PC-3M细胞的端粒醇活性分别增高0.18、1.25、2.89和6.42倍。结论 GM-CSF能够显著增强PC-3M细胞端粒酶的活性,GM-CSF这种功能可能是其促进前列腺癌细胞增殖的机制之一。

Abstract:Objective To study the possible association between granulocyte-macrophage colony stimulating factor (GM-CSF) and the telomerase activity of human prostate cancer cell line PC-3M. Methods PC-3M cell line was cultured with GM-CSF in the concentrations of 10 ng/ml, 20 ng/ml, 40 ng/ml and 100 ng/ml, respectively. Telomerase activity was measured qualitatively and quantitatively by telomerase PCR ELISA method. Results The telomerase activity was elevated in PC-3M when incubated with GM-CSF in a concentration-dependent manner. The relative telomerase activity was increased by 0.18, 1.25, 2.89 and 6.42 times by GM-CSF of 10 ng/ml, 20 ng/ml, 40 ng/ml and 100 ng/ml correspondingly. Conclusion The telomerase activity could be elevated by GM-CSF, which might be one of the proliferation mechanism of advanced prostate cancer cells.
