摘    要:目的研究自组装肽包裹嗅神经鞘细胞(OECs)移植对大鼠脊髓损伤(SCI)后轴突再生的影响。方法成年雄性SD大鼠40只,行脊髓T_(10)节段背侧横断术,动物随机分为4组,每组10只。SAP-OECs组于间隙内移植SAP包裹的OECs悬液,SAp、OECs对照组同法分别移植等量SAP及DF12-OECs悬液;单纯全切对照组不做处理。术后2或4周过量麻醉处死动物,脊髓损伤节段连续冰冻矢状切片。行抗GAP-43和GFAP免疫组织化学和免疫荧光双标染色。结果SAP-0ECs组损伤区内有大量GAP-43免疫反应纤维,SAP组相较OECs及单纯半切对照组则可见相当数量GAP-43免疫反应纤维。OECs及单纯半切对照组损伤区周围可见一条致密度的GFAP阳性的反应性星形胶质细胞条带,而SAP组GFAP免疫反应纤维比对照组少,且多于SAP-OECs组。此外激光共聚焦显微镜下观察SAP-OECs组在损伤后4周GAP-43免疫反应纤维能通过位于脊髓损伤区尾端的GFAP阳性区域。结论自组装肽包裹嗅鞘细胞可以抑制脊髓损伤后胶质疤痕形成并促进轴突再生。
