双环醇对肝硬化模型大鼠炎性因子、胃肠功能及肝功能的影响

摘    要:目的] 探讨双环醇对肝硬化模型大鼠炎性因子、胃肠功能及肝功能的影响.方法] 选择雄性SD大鼠60只并分为对照组、模型组以及双环醇低剂量组、中剂量组、高剂量组,采用四氯化碳皮下注射的方式建立肝硬化模型.双环醇低剂量组、中剂量组、高剂量组分别给予0.1 g/kg、0.2 g/kg、0.3 g/kg双环醇灌胃.干预后8周时,检测并比较各组小鼠炎性因子白细胞介素-1β(IL-1β)、白细胞介素-8(IL-8)、肿瘤坏死因子-α(TNF-α)] 、胃肠黏膜屏障功能指标降钙素原(PCT)、D-乳酸、内毒素(LPS)] 、肝功能指标丙氨酸氨基转氨酶(ALT)、天冬氨酸氨基转移酶(AST)] 的变化.结果] 干预后8周时,模型组大鼠血清中IL-1β、IL-8、TNF-α、ALT、AST、PCT、LPS、D-乳酸水平均显著高于对照组(P<0.05);双环醇低剂量组、中剂量组、高剂量组大鼠血清中IL-1β、IL-8、TNF-α、ALT、AST、PCT、LPS、D-乳酸水平均低于模型组(P<0.05);双环醇剂量越大,血清中IL-1β、IL-8、TNF-α、ALT、AST、PCT、LPS、D-乳酸的水平越低(P<0.05).结论] 双环醇对肝硬化模型大鼠的胃肠功能及肝功能具有保护作用,同时能够抑制炎性因子的合成,是治疗肝硬化的理想药物.

Effect of Double-loop Alcohol on Inflammatory Factors,Gastrointestinal Function and Liver Function in Rats with Liver Cirrhosis
Abstract:Objective]To study the effect of double loop alcohol on inflammatory factors, gastrointestinal function and liver function in rats with liver cirrhosis.Methods]Sixty male SD rats were selected and randomly divided into control group, model group and bicyclol low dose group, middle dose group, high dose group.The liver cirrhosis model was established by subcutaneous injection of carbon tetrachloride.Bicyclol low dose, middle dose and high dose groups were given 0.1g/kg and 0.2g/kg, 0.3 g/kg of bicyclol orally.At 8 weeks after intervention, serum was collected and serum levels of inflammatory factors (IL-1, IL-8, TNF-α), gastrointestinal mucosal barrier function indexes (PCT, LPS, D-lactate) and liver function indexes (ALT, AST) were determined.Results]At 8 weeks after the intervention, serum IL-1, IL-8, TNF-α, ALT, AST, PCT, LPS, D-lactate contents of rats in the model group were significantly higher than those in the control group (t=13.729~23.179, P<0.05).Serum IL-1, IL-8, TNF-α, ALT, AST, PCT, LPS, D-lactate contents of rats in bicyclol low dose, middle dose and high dose groups were significantly lower than those of model group (t=6.324~20.938,P<0.05).As bicyclol dose increased, serum IL-1, IL-8, TNF-α, ALT, AST, PCT, LPS, D-lactate contents decreased (t=2.224~7.456,P<0.05).Conclusion]Double-loop alcohol has protective effect on the gastrointestinal function and liver function of liver cirrhosis model rats, and can inhibit the synthesis of inflammatory factors, which is an ideal drug for the treatment of liver cirrhosis.
