Syndecan-1和HPA-1 mRNA在胃癌组织中的表达及其与胃癌侵袭转移的关系

摘    要:背景与目的:Syndecan-1和HPA-1(乙酰肝素酶-1)可能参与多种恶性肿瘤的侵袭转移,但关于两者与胃癌的关系较少报道。本研究旨在探讨Syndecan-1mRNA和HPA-1mRNA在胃癌组织中的表达及两者与胃癌侵袭转移的关系。方法:采用实时荧光定量逆转录聚合酶链反应(RT-PCR)分别检测了58例胃癌及58例配对的癌旁组织(距癌2cm)和58例配对远离胃癌的手术切缘正常组织(距癌5cm以上)Syndecan-1和HPA-1的mRNA的表达水平,分析探讨两者与胃癌临床病理特征和胃癌侵袭转移的关系。结果:HPA-1mRNA在胃癌中表达的阳性率(86.2%)显著高于癌旁组织(27.6%)和正常组织(5.2%)(P均〈0.001),癌旁组织明显高于正常组织(P均〈0.05)。Syndecan-1mRNA在正常组织中表达的阳性率(98.3%)显著高于癌旁组织(25.9%)和胃癌组(5.2%)(P均〈0.001),癌旁组织中明显高于胃癌组织(P均〈0.05)。Syndecan-1的表达减弱和HPA-1的表达增强与肿瘤的分化程度、浸润深度、淋巴结转移、远处转移及TNM分期显著相关(P〈0.05)。Syndecan-1mRNA的表达与HPA-1mRNA的表达呈负相关。结论:Syndecan-1mRNA在正常胃组织中呈高表达,HPA-1mRNA在胃癌组织中呈高表达,且Syndecan-1的表达减弱与HPA-1的表达增强可促进胃癌的侵袭转移。联合检测Syndecan-1mRNA及HPA-1mRNA对于指导胃癌临床治疗及判断预后有重要价值。
