TGF-β1对人早孕滋养层细胞和绒毛膜癌JAR细胞增殖的影响及其意义

目的:研究转化生长因子-β1(TGF-β1)对人早孕滋养层细胞和人绒毛膜癌JAR细胞增殖的影响,探讨TGF-β1在人妊娠滋养细胞疾病发病机制中的作用和意义。方法:分别用细胞计数法、MTT比色法研究TGF-β1对两种细胞增殖活性的影响。结果:随着TGF-β1浓度的增高,TGF-β1抑制人正常早孕滋养层细胞的增殖作用加大;在一定的浓度范围内,以浓度依赖性和时间依赖性的方式促进JAR细胞的增殖,提示TGF-β1在良性滋养细胞疾病的恶变中可能有促进作用。结论:TGF-β1具双重作用,既抑制人正常早孕滋养层细胞的增殖,又在一定浓度范围内以浓度依赖性和时间依赖性的方式促进恶性滋养细胞JAR细胞系的增殖。

Effect and significance of TGF-β1 on cell proliferation of human first trimester cytotrophoblasts and JAR cells

Objective:To explore the role and mechanism of transforming growth factor-β1(TGF-β1) in the proliferation of human first trimester cytotrophoblasts and JAR cells.To explore the possible role and significance of TGF-β1 in human gestational trophoblastic disease(GTD).Methods:Cytometry and MTT were used respectively to detect the effect of TGF-β1 on cell proliferation activity of human first trimester cytotrophoblasts and JAR cells.Results:The growth and proliferation of human first trimester cytotrophoblasts were inhibited significantly by TGF-β1 in the concentration and time dependent manners compared with those of the control after the human first trimester cytotrophoblasts were treated with 0 ng/ml,0.01 ng/ml,0.1 ng/ml,1 ng/ml,10 ng/ml and 100ng/ml TGF-β1 for 24 h,48 h and 72 h.While within some certain concentration range,proliferation of JAR cells were significantly stimulated by the treatment of TGF-β1 in the concentration and time dependent manners in vitro.The study showed that the possibly promoting effect of TGF-β1 in benign trophoblastic disease.Conclusion:TGF-β1 has dual effects: on one hand,it can inhibit the proliferation capability of human first trimester cytotrophoblasts;on the other hand,within some certain concentration range,it can stimulate the proliferation capability of JAR cells by concentration and time dependence manners.
