摘    要:目的 研究去整合素金属蛋白酶-12(ADAM-12)和增殖细胞核抗原(PCNA)在膀胱癌中的表达,并探讨其表达与膀胱癌发病之间的关系.方法 应用免疫组织化学的方法 检测43例膀胱癌、15例正常膀胱组织中ADAM-12及PCNA的表达情况,并分析其与膀胱癌患者临床特征及病理分期的关系.结果免疫染色标本中,ADAM-12在膀胱癌组织中的表达明显高于正常膀胱组织,两者比较差异有统计学意义(x2=11.241,P=0.010).病理分期高的膀胱癌组织表达明显高于分期低者(x2=22.368,P<0.001).PCNA在膀胱癌中的表达明显高于正常膀胱组织,两者比较差异有统计学意义(x2=9.250,P=0.026).病理分期高的膀胱癌组织的表达明显高于分期低者(x2=10.665,P=0.014).ADAM-12与PCNA在膀胱癌中的表达呈正相关(r=0.997,P<0.001).结论 膀胱癌组织中ADAM-12蛋白和PCNA呈过表达,可能与肿瘤的发生发展密切相关;ADAM-12是膀胱癌具有潜在价值的肿瘤标记物,可以成为一项新的临床观察指标.

Abstract:Objective To investigate the expression of adamalysin-12 (ADAM-12) and PCNA in human bladder carcinoma and to investigate their correlation with different grades and stages of bladder cancer.Methods Biopsies of 15 normal bladder and 43 bladder tumors were analyzed.Immunohistochemistry was conducted to detect the expression of ADAM12 and PCNA in the biopsies.Results Postive expression signals of ADAM12 were detected significantly higher in the bladder cancer biopsies than that in the normal ones (P = 0.010).Those with lower histological grade had a higher expression level of ADAM-12 compared to the higher histological grades (P <0.001 ).Positive expression signals of PCNA were detected significantly higher in the bladder cancer biopsies than that in the normal ones (P = 0.026).Those with lower histological grade had a higher expression level of PCNA (P =0.014).There was a positive correlation between the expression of ADAM-12 and PCNA in bladder cancer (r =0.997,P < 0.001 ).Conclusions The overexpression of ADAM-12 and PCNA in the biopsies of bladder tumors shows that protein expression of ADAM-12 and PCNA correlated with tumor stage and grade.Furthermore,ADAM-12 may be a promising biomarker of bladder cancer in the clinical implication.
